From Shinjuku Station, take the Shinjuku Line to Ichigaya Station, then switch to the Yurakucho Line train — you can get off at Toyosu Station, from there it's a 10-minute walk; or catch the Yurikamome Line one stop over.

In general, Koto Ward is an old traditional neighborhood (although most things turned into ashes during WW2). Kiyosumi, Fukagawa, Monzen-Nakacho sounds very old-fashioned grannies' places to the local Japanese, although I personally think those areas are quite nice and relaxing neighborhoods good for a long-term stay.
